We designed primers forward and reverse overlapping the restriction site, both of them with a single mismatch on the second T of the site "CATATG".
Primer forward: 5’ – CCGCATAAGGTGCACTCTCAGTACAAT – 3’
Primer reverse: 5’ – GTGCACCTTATGCGGTGTGAAATACCG – 3’
After the mutagenesis, the restriction site was changed from CATATG to CATAAG.
